Hi,

I was referred by someone inside to a Big 4 about 4 weeks
ago and haven't received a call or an email from them.
Is it safe to assume my chances to interview with them
are very unlikely? I often wonder if it had something to
do with me not submitting a cover letter during the
application process...

I just wanted to get some idea so I can move on to
applying other companies. Please advice.

Cover letter doesn't matter, it's all about your experience.

I also had my resume submitted by a Big4 tax director and haven't heard from the firm, only for the recruiter to tell me that it is on file should this director's group have needs.

I also heard from another Big 4 recruiter that even if a partner submits your resume and you don't have the goods (for example low GPA), they still will not give you an interview.

While it helps for someone in the firm to submit your resume, I think HR still has to decide whether to give you an interview and another reason they don't is because there are currently no open positions.

I applied for an internal audit position. My GPA is not
low. I have all the necessary qualifications. At this
point, I am not going to wait up but to move onto applying
other big 4.

You are done.  If they were remotely interested at your resume, then they would have phone screened you and presented it to a hiring manager.  From there, that will decide whether or not you will get an interview.  No phone screen? No interest.
